Nucleic acid testing (NAT) is a molecular technique for screening blood donations to reduce the risk of transfusion transmitted infections (TTIs) in the recipients, thus providing an additional layer of blood safety. NAT technique is highly sensitive and specific for viral nucleic acids. It is based on amplification of targeted regions of viral ribonucleic acid or deoxyribonucleic acid (DNA) and detects them earlier than the other screening methods thus, narrowing the window period of transfusion transmitted infections.
Flow cytometry is a versatile technique used in biomedical research and clinical diagnostics to analyze and quantify cells and particles in a fluid suspension. It involves passing cells or particles through a laser beam, measuring their characteristics such as size, granularity, and fluorescence. The complement-dependent cytotoxicity crossmatch (CDC-XM), a technique that uses T and B lymphocytes to detect donor-specific antibodies (DSA) via activation of the classical pathway of complement, has been widely used for the detection of alloantibodies in transplant patients since it was introduced in the 1960s. CDC crossmatch is still considered to be the gold standard test to predict hyperacute graft rejection
